Prajnesh, who qualified into the tournament and stunned world number 23 Denis Shapovalov in the opening round, lost to Guido Pella 6-7(4) 4-6.
Prajnesh will lock horns with top American Taylor Fritz, who knocked out India's Vijay Sundar Prashanth.
Ramkumar, now India's number one singles player at 156, faces seasoned Frenchman Paul-Henri Mathieu, who is now placed 213 in the ATP ranking chart.
The 22-year-old from Chennai is now India's best-ranked player in the singles.
